Some criteria for choosing a suitable internal standard:
- The internal standard must have the same chemical, physical similar properties as that of the analyte, commonly, they’re analogs, homologs, isotopes, .. so it will have the same behaviors like the analyte during clean-up, sample procedure, ...
- Stable, high purity and not interfere with the analyte, don’t interact chemically or physically with analyte, matrix sample, and mobile phases.
- The similiar detector response to that of the analyte.
- The internal standard mustn’t overlap with the sample peak and elute near the peak of analyte.
- The internal standard must exists with very low concentration in the sample that its initial signal can’t be detected, ideally it shouldn’t be present naturally in the matrix sample.
- Commercially available.
